Yep! The circular indentation indicates the orientation. Top left. Some ICs also have their own notch in the top middle (like the outline on the PCB). It’s also common practice to add 8 pin sockets instead of soldering the ICs directly. Helps prevent situations like this...

UV templates are complex enough to me that I would be hesitant to not use a computer to make them. My suggestion would be to get creative with manual artwork. A lot of folks have made some beautiful pedals with spray paint and paint markers.
