I was a moron and put the headers on backwards, I don't think it will make a difference but could someone let me know if it would?

I am working on troubleshooting it right now, third one I have built and this one is not passing signal. Also, the LED on V1 is very dim.

Putting the headers in backwards will absolutely not work. You'll need to pull those and fix it.
 
Nothing is jumping out from the picture, definitely look for shorts, look for cold solder joints. It's hard to tell for sure as it could just be lighting, but the solder joint on the yellow input jack wire looks suspect.
 
Where are you checking for a signal, at the XLR or the 1/4" out jack ?
You should be able to check the XLR output by putting a 1/4" jack there instead of the transformer board.
If you check the XLR try with different tubes in V1 and no tube in V2.
That way you are testing the first half of the circuit only, it helps establishing where the fault is.
