If your guitar isn't set up, in tune, etc. Your pedals are going to sound like shit anyway.

Pickup height is critical! I recently bought a Gibson ES-330, which is a fantastic guitar. (I got a crazy price so had to pounce!) But for a rather pricey guitar there is no way to adjust the height of the pickups. So I had one shim left from a pack I got from Lollar a few years back and used that for the bridge pickup. It wasn't quite enough so I went down to the local art supplies store and bought some thick black card and cut some more. Now the pickups are nice and close to the strings (P90s can be quite close without causing problems) and the guitar sounds better than ever.

Filter'trons need to be quite close too, but traditional Strat pickups need a lot of room unless you want the warble.

I've yet to find a tuner pedal that does what it actually says it's going to do.

I hear no difference, on or off.

I've played tuned guitars, I still sounded terrible.
See it this way.

If you don’t tune, « it » sounds terrible.
If you do, you sound terrible.

IMO, rather stay in doubt in that case.
