I thought I had ordered an A250k pot but apparently I did not. I have an A100k an A500k, could I sub either one?

Either one should be fine in place of the A250k volume pot, the actual value of that pot isn't critical.

You could also run a 470k or 510k resistor connecting pin 1 and 3 on the 500kA pot and it will get you in the ballpark of a 250kA pot.

One last question if you don't put the jumper for the true bypass the effect will not work. Looking at the schematic this is required and most likely why I am only getting a boost and no tremolo.

You need the jumper or no tremolo just confirmed. Used an A100k and it works fine.
